Si valuta il potenziale utilizzo dei microrganismi in numerosi campi di applicazione tra cui il biorisanamento di matrici contaminate e il trattamento biologico di acque reflue. Questo approccio consente l\u2019identificazione e la caratterizzazione dei microrganismi in sistemi ambientali complessi e delle loro potenzialit\u00e0 metaboliche utilizzabili in tecnologie ecosostenibili volte alla mitigazione del rischio per l\u2019ambiente e la salute umana. Le attivit\u00e0 sono mirate allo studio di microorganismi coinvolti in un ampio spettro di processi ambientali quali il biorisanamento di matrici contaminate, la digestione anaerobica, il trattamento biologico di acque reflue e la valorizzazione di scarti organici per la produzione di biogas, biopolimeri e sostanze ad alto valore aggiunto. Inoltre, si svolgono studi di interesse igienico-sanitario che prevedono il monitoraggio di organismi patogeni e di geni resistenti agli antibiotici in diverse matrici ambientali.<br \/>\nL\u2019apparato strumentale a disposizione consente di eseguire servizi di:<\/p>\n<ul>\n<li style=\"text-align: justify;\">identificazione e quantificazione di microorganismi in matrici ambientali mediante analisi microscopiche ad epifluorescenza;<\/li>\n<li style=\"text-align: justify;\">quantificazione della presenza e dell\u2019attivit\u00e0 di microorganismi di interesse biotecnologico e dei relativi geni (16S rRNA e geni funzionali mediante analisi qPCR,RT-qPCR e Droplet Digital PCR);<\/li>\n<li style=\"text-align: justify;\">caratterizzazione genomica e metagenomica di comunit\u00e0 microbiche complesse mediante Next Generation Sequencing (NGS) mirato ai geni 16S rRNA di Batteri ed Archaea e 18S rRNA di Eucarioti e Whole Genome Shotgun Sequencing (WGS);<\/li>\n<li style=\"text-align: justify;\">determinazione di microrganismi di interesse igienico-sanitario mediante analisi con Droplet Digital PCR.<\/li>\n<\/ul>\n<p>In aggiunta alla caratterizzazione tassonomica e funzionale di comunit\u00e0 microbiche miste, sia naturali che selezionate in sistemi di interesse biotecnologico, siamo in grado di offrire le seguenti analisi specialistiche:<\/p>\n<ul style=\"text-align: justify;\">\n<li>analisi di carattere igienico-sanitario (determinazione di cisti di Giardia ed oocisti di ryptosporidium); monitoraggio di microrganismi patogeni (Salmonella spp., Legionella pneumophila, Pseudomonas aeruginosa) tramite Loop-mediated isothermal Amplification (LAMP); analisi di geni di resistenza agli antibiotici (geni ampC, mecA, ermB, sul1, sul2, tetA, tetO, tetW, vanA) mediante qPCR;<\/li>\n<li>analisi di biofilm microbici in sistemi naturali, inclusa la plastisfera, e artificiali, inclusi i sistemi industriali, mediante tecniche di identificazione in situ (FISH-CARD-FISH) combinate con la microscopia confocale a scansione laser (CLSM);<\/li>\n<li>analisi di batteri arsenito ossidanti e arseniato riducenti in sistemi di trattamento per le acque potabili mediante qPCR (geni aioA, arsBC, arrA, arxA);<\/li>\n<li>analisi di gruppi microbici funzionali responsabili dei processi di digestione anaerobica mediante qPCR: produzione di metano (gene mcrA); produzione di biopolimeri (PHA, analisi delle PHA sintasi); produzione di acidi grassi a lunga catena (analisi di lattato-deidrogenasi e alcol-deidrogenasi).<\/li>\n<\/ul>\n<p>&nbsp;<\/p>\n<p><strong>Valorizzazione della Ricerca &#8211; Spin Off CNR\u00a0<em>&#8220;<a href=\"https:\/\/trireme.it\/\" target=\"_blank\" rel=\"noopener\">TRIREME<\/a>&#8220;<\/em><\/strong><\/p>\n<p style=\"text-align: justify;\">Il laboratorio di Microbiologia Ambientale e Biologia Molecolare ospita lo Spin Off \u201c<a href=\"https:\/\/trireme.it\/\" target=\"_blank\" rel=\"noopener\">Trireme<\/a> \u2013 Technologies for Reliable and Innovative REMEdiation\u201d. <a href=\"https:\/\/trireme.it\/\" target=\"_blank\" rel=\"noopener\">TRIREME<\/a> offre servizi di monitoraggio ambientale e caratterizzazione chimico-fisica e biomolecolare, progettazione, messa in sicurezza, bonifica e ripristino ambientale dei siti inquinati mediante tecnologie innovative. Nell\u2019ambito delle attivit\u00e0 in convenzione con IRSA-CNR si eseguono analisi biomolecolari quantitative (qPCR, RT-qPCR) di biomarcatori in matrici ambientali inquinate, ivi inclusi Dehalococcoides mccartyi e geni funzionali per la declorazione riduttiva (pceA, tceA, bvcA, vcrA), solfato-riduzione (dsrAB, apsA), ossidazione di inquinanti organici quali ad esempio bifenildiossigenasi (bphA) e monossigenasi (etnE, etnC). Il principale ambito di ricerca \u00e8 l\u2019identificazione di fonti di contaminazione antropica, in particolar modo correlata alle tecniche agronomiche, per la tutela delle acque e dei suoli da contaminazione dei composti chimici di fertilizzazione (nitrati, fitofarmaci e glifosato). Le attivit\u00e0 attualmente operative mirano all\u2019identificazione dei principali microrganismi presenti nelle matrici ambientali, all\u2019identificazione e studio del loro sviluppo in correlazione alla presenza di composti chimici che ne limitano o modificano l\u2019accrescimento e\/o la presenza.<\/p>\n<p style=\"text-align: justify;\">Attraverso lo sviluppo di algoritmi matematici e modelli il gruppo di ricerca attua uno studio sull\u2019andamento spaziale e temporale ipotizzando le possibili cause delle diversificazioni individuate e individuando cos\u00ec possibili
